KE00035 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The MHC class Ⅰ ELISA Kit is to be used to detect and quantify protein levels of endogenous MHC class Ⅰ molecules. The assay recognizes human MHC class Ⅰ molecules. A polyclonal MHC class I antigen A*3 antibody has been pre-coated onto the microwells. The MHC class Ⅰ proteins in samples is captured by the coated antibody after incubation. Following extensive washing, a monoclonal MHC class I antigen A antibody is added to detect the captured MHC class Ⅰ proteins. For signal development, horseradish peroxidase (HRP)-conjugated Anti-mouse antibody is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450nm.

Background

Major histocompatibility complex (MHC) antigens, also know in human as human leukocyte antigens (HLA), are encoded by genes located on the short arm of chromosome 6 (6p21.3). MHC class I molecules (HLA-A, -B and -C) are polymorphic membrane glycoproteins composed of two polypeptide chains, a heavy alpha chain encoded by variant HLA-A, -B or -C gene and a smaller beta chain (β2-microglobulin). They are peptide-binding proteins that present processed antigens to T lymphocytes.
