  • Purity of recombinant human FGF-19 was determined by SDS- polyacrylamide gel electrophoresis. The protein was resolved in an SDS- polyacrylamide gel in reducing and non-reducing conditions and stained using Coomassie blue.

  • Recombinant human FGF-19 (HZ-1330) stimulates dose-dependent proliferation of the NIH/3T3 mouse fibroblast cell line. Viable cell number was quantitatively assessed by Prestoblue Cell Viability Reagent. NIH/3T3 cells were serum starved with 0.02% FBS during treatment with increasing concentrations of recombinant human FGF-19 for 72hrs in defined medium. The EC50 was determined using a 4- parameter non-linear regression model. The EC50 values range from 50-300 ng/mL.

Reconstitution Briefly centrifuge the vial before opening. It is recommended to reconstitute the protein to 0.2 mg/mL in sterile 1x PBS pH 7.4 containing 0.1% endotoxin-free recombinant human serum albumin (HSA). Gently swirl or tap vial to mix.

Background

FGF-19 is a member of the endocrine subfamily of FGFs. It primarily binds and activates signaling pathways downstream of FGFR4 in the liver. Through downregulation of the CYP7A1 enzyme, FGF-19 can prevent overproduction of bile salts in the liver and contribute towards their homeostasis. It is also a regulator of glucose and lipid metabolism alongside FGF-21. FGF-19 can additionally stimulate hepatocyte proliferation and has been correlated with the progression of hepatocellular carcinoma when its signaling levels are increased.
