KE00295 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The B7-H3 (CD276) ELISA kit is to be used to detect and quantify protein levels of endogenous B7-H3 (CD276). The assay recognizes human B7-H3 (CD276). An antibody specific for B7-H3 (CD276) has been pre-coated onto the microwells. The B7-H3 (CD276) protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for B7-H3 (CD276) is added to detect the captured B7-H3 (CD276) protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
B7-H3 (CD276) is a type I transmembrane protein expressed on many tissues and cell types. B7-H3 is a 100-kDa glycoprotein that belongs to the B7 immunoregulatory family and participates in the regulation of T-cell-mediated immune response probably by functioning as both a T cell costimulator and coinhibitor. Overexpressed on a wide range of human solid cancers, B7-H3 has been implicated in cancer progression and metastasis and becomes an attractive target for cancer immunotherapy. In addition to the membrane form, B7-H3 can also exist as a soluble form (sB7-H3) generated either through alternative splicing or cleavage from the cell surface, and the serum level of sB7-H3 has been correlated with prognosis in various malignancies.
