Product Information
KE00026 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The CD3E ELISA kit is to be used to detect and quantify protein levels of endogenous CD3E. The assay recognizes human CD3E. An antibody specific for CD3E has been pre-coated onto the microwells. The CD3E protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody specific for human CD3E is added to detect the captured human CD3E protein. For signal development, HRP-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
CD3 is a complex of proteins that directly associates with the T cell receptor (TCR). The TCR/CD3 complex of T-lymphocytes consists of either a TCR alpha/beta or TCR gamma/delta heterodimer coexpressed at the cell surface with the invariant subunits of CD3 labeled gamma, delta, epsilon, and zeta. The TCR recognizes antigens bound to major histocompatibility complex (MHC) molecules. TCR-mediated peptide-MHC recognition is transmitted to the CD3 complex, leading to the intracellular signal transduction. CD3E encodes the CD3-epsilon polypeptide which plays an essential role in T-cell development in addition to its role of signal transduction in T-cell activation. Defects in CD3E cause immunodeficiency.
