KE00344 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The human Cyclin D1 ELISA kit is to be used to detect and quantify protein levels of endogenous human Cyclin D1. The assay recognizes human Cyclin D1. An antibody specific for human Cyclin D1 has been pre-coated onto the microwells. The human Cyclin D1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human Cyclin D1 is added to detect the captured human Cyclin D1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

CCND1 (cyclin D1), also known as PRAD1 or BCL1, belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance throughout the cell cycle. Cyclin D1 is mostly known for the role played in the nucleus, as regulator of cell cycle progression. Cyclin D1 modulates the transition from G1 to S phase through its action as allosteric regulator of the cyclin-dependent kinase 4 (CDK) and CDK6 (PMID: 33317149). The active Cyclin D1/CDK4 complex translocates into the nucleus, phosphorylates retinoblastoma (RB) protein together with Cyclin E/CDK2, and dismisses the repressive action on the E2F transcription factor, which regulates the transcription of specific genes required for cell proliferation (PMID: 8449399, PMID: 9447971). Dysregulation of Cyclin D1 transcription, accumulation and ubiquitination, as well as assembly and hyperactivation of its cognate CDK, lead to uncontrolled cell growth. Thus, Cyclin D1 is regarded as an oncogenic driver in different types of cancers including breast cancer, lung cancer, and melanoma (PMID: 31885322). Cyclin D1 is expressed by the majority of normal human cells apart from cells derived from bone marrow stem cell lines (PMID: 1386335).
