KE10138 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse CD34 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse CD34. The assay recognizes mouse CD34. An antibody specific for mouse CD34 has been pre-coated onto the microwells. The mouse CD34 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for mouse CD34 is added to detect the captured mouse CD34 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

CD34 is a 105- to 120-kDa transmembrane phosphoglycoprotein consisting of a cytoplasmic tail that contains PDZ-domain binding motifs, a single transmembrane helix and an extracellular domain that is heavily sialylated, O-linked glycosylated, and contains some N-linked glycosylation sites. It is expressed by the majority of hematopoietic stem/progenitor cells and also by a multitude of other nonhematopoietic cell types including muscle satellite cells, corneal keratocytes, interstitial cells, epithelial progenitors, and vascular endothelial cells. CD34 is widely used as a marker of vascular endothelial cells and hematopoietic stem/progenitor cells and mediates cell adhesion and lymphocyte homing by binding L-selectin and E-selectin ligands.
