Human peripheral blood mononuclear cells (0.5 x 10^6 cells/mL) were cultured in RPMI supplemented with 10% fetal bovine serum, 5 μg/mL PHA, and 10 ng/mL recombinant human IL-2. After 7 days of culture, cells were changed into DMEM with 10% fetal bovine serum, 10 ng/mL PMA, 1 μg/mL Ca2+ ionophore, and 5 μg/mL PHA. Cell culture supernate was collected after 1d of stimulation and measured 292.5 pg/mL in stimulated PBMC supernatant, 11.2 pg/mL in unstimulated PBMC supernatant.

Serum of sixteen individual healthy human donors was measured. The human IL-4 concentration of detected samples was determined to be 7.97 ng/mL with a range of 2.25 - 19.49 ng/mL.
