Understanding the role of non-structural proteins (NSPs) and their implication in long-term severe repercussions of SARS-CoV-2 infection
Since the onset of the COVID-19 pandemic, research into coronaviruses; specifically SARS-CoV-2, has increased exponentially. Study of the viral spike protein, and targets such as ACE2 and TRPMSS2 have been some of the most popular research topics. Understanding the role of non-structural proteins (NSPs) and their implication in long-term severe repercussions of SARS-CoV-2 infection is an important step in the understanding of severe disease symptoms.
NSPs aid the SARS-CoV-2 virus in establishing the replication and transcription complex (RTC) through endoplasmic reticulum (ER) hijacking and immune evasion. RTC along with membrane rearrangement can cause organelle damage such as ER stress, changes to autophagy machinery, Golgi fragmentation, and lysosome membrane alteration. This is thought to play a leading role in some of the more severe symptoms of SARS-CoV-2 infection such as neuronal, lung, and other organ damage.
