  • Purity of GMP-grade recombinant human HGF was determined by SDS- polyacrylamide gel electrophoresis. The protein was resolved in an SDS- polyacrylamide gel in reducing and non-reducing conditions and stained using Coomassie blue.

Viable cell number was quantitiatively assessed by PrestoBlue Cell Viability Reagent. 4MBr-5 cells were treated with increasing concentrations of recombinant human HGF for 120 hours. The EC50 was determined using a 4- parameter non-linear regression model. Activity determination was conducted in triplicate on a validated bioassay. The EC50 value ranges from 5-25 ng/mL.

Reconstitution Briefly centrifuge the vial before opening. It is recommended to reconstitute the protein to 0.2 mg/mL in sterile 1x PBS pH 7.4 containing 0.1% endotoxin-free recombinant human serum albumin (HSA). Gently swirl or tap vial to mix.

Background

Hepatocyte growth factor (HGF) is the most potent mitogen of mature hepatocytes in primary culture. It is secreted by mesenchymal cells and acts as a multi-functional cytokine on cells of mainly epithelial origin (PMID 1530787, 2531289). HGF exerts a variety of effects on several cells, such as inflammation, tissue repair, morphogenesis, angiogenesis, tumour propagation, immunomodulation of viral infections and cardio-metabolic activities (PMID: 1846706; 15070743; 1383237; 1827664).
