Background

IL-22 is a 6-α-helix, bundle cytokine that is a member of the IL-10 family of cytokines. It is primarily produced by innate lymphoid cells (ILCS) and multiple T cell subsets including αβT, γδT, and NKT cells. Its production is regulated upstream by several different cytokines including IL-23, IL-1β, and IL-7. IL-22 binds with the IL-22 receptor, expressed on stromal and epithelial cells in a wide range of tissues, but it can also bind in circulation with the IL-22 binding protein (IL-22BP), which functions as a negative regulator. Binding with the receptor triggers several downstream pathways resulting in pro-inflammatory responses, increased mucus production, as well as increased production of anti-bacterial and anti-fungal proteins. IL-22 also plays a role in promoting wound healing and tissue regeneration in response to injury. Overexpression of IL-22 has been linked to several cancers, most notably colon cancer (PMID: 25706098, 33042126).
