Cancer is still a leading cause of mortality. Each year 18 million people worldwide get diagnosed with cancer. The current trend is increasing. This makes cancer research more important.
Broader knowledge of metastasis, proliferative signaling, apoptosis, autophagy, metabolism, or inflammation and protein abnormalities is the main focus of research nowadays. Antibodies and Nanobodies are important tools in this field.
ChromoTek’s research tools can help to understand cancer cell signaling pathways and cellular processes, as a complex system machinery. Here is an overview of our Nanobody-based tools for cancer research:
P53-C-term-Trap & P53-N-term-Trap
The tumor suppressor protein p53 plays a major role in cell cycle control, apoptosis, and senescence.
ChromoTek offers two different p53-Traps for immunoprecipitation of the p53 proteome with complementary p53-N-term & p53-C-term Traps:
- Pull down of human p53 isoforms alpha, beta, and gamma with p53 N-term-Trap. The p53 N-term-Trap binds to epitope aa 1-81.
- Pull down of human and mice p53 isoforms alpha with p53 C-term-Trap. The p53 C-term-Trap binds to epitope aa 302-393.
MK2-Trap
MK2 (MAP kinase-activated protein kinase 2, MAPKAP-K2) is a stress-activated serine/threonine-protein kinase involved in cytokine production, endocytosis, reorganization of the cytoskeleton, cell migration, cell cycle control, chromatin remodeling, DNA damage response, and transcriptional regulation.
ChromoTek’s MK2-Trap Agarose contains anti-MK2 Nanobody conjugated beads for immunoprecipitation of MK2-fusion proteins from human, mouse, and hamster. The MK2-Trap recognizes unphosphorylated MK2 and Phospho-MK2 (Thr222). Specificity on Phospho-MK2 (Thr334) is not tested.
Mdm4/HdmX
Double minute 4 protein (Mdm4/HdmX) plays a role in cell cycle control and apoptosis: It inhibits p53/TP53- and TP73/p73-mediated cell cycle arrest and apoptosis by binding its transcriptional activation domain. Furthermore, it inhibits degradation of MDM2 and can reverse MDM2-targeted degradation of TP53 while maintaining suppression of TP53 transactivation and apoptotic functions.
Pull down of human Mdm4/HdmX (epitope: aa 1-129) with Mdm4/HdmX-Trap. Please note that binding of the Mdm4/HdmX-Trap interferes with binding of p53 to Mdm4/HdmX.
PARP1
Poly (ADP-ribose) polymerase 1 (PARP1) is one of the most abundant proteins in the nucleus and is involved in many cellular processes including DNA repair, transcriptional regulation, and modulation of chromatin structure.
The PARP1-Trap contains affinity beads for immunoprecipitation of PARP1 consisting of an anti-PARP1 Nanobody/VHH conjugated to agarose.
Vimentin-Label & Vimentin-Chromobody
Vimentin is an intermediate filament protein. The vimentin filament network plays a key role in cell architecture and signaling in mesenchymal cells. Vimentin is also an important marker of the epithelial–mesenchymal transition, a highly dynamic cellular process involved in the initiation of metastasis and cancer progression.
The Vimentin-Label is an anti-Vimentin Nanobody conjugated to ATTO 488 for IF/microscopy. The Vimentin-Chromobody® is a fluorescent probe to analyze endogenous vimentin in a physiological context, for live cell imaging and to trace epithelial–mesenchymal transition in real-time.
GFP
Obviously, green fluorescent protein (GFP) is not an oncogene. Nevertheless, as one of the most frequently used protein tags, GFP plays an important role in cancer research. The extraordinary performance and specificity of the GFP-Trap for immunoprecipitation of GFP-tagged proteins results in reliable data. The GFP-Trap is cited in more than 50 publications in cancer research. Please contact us with any questions.
