Ionomycin is a calcium ionophore and an antibiotic produced by Streptomyces conglobatus. The divalent cation selectivity order for ionomycin was Ca > Mg >> Sr = Ba[1]. Ionomycin at concentrations of 1-5μM induced the proliferation of highly purified human T cells in the presence of monocytes, associated with enhanced IL-2R expression and IL-2 synthesis. Ionomycin at 2μM induced the hydrolysis of phosphoinositides in purified T cells. Treatment of T cells with ionomycin (2μM) also induced the activation of protein kinase C[2]. Ionomycin at a concentration of 10µmol/L activated mouse oocytes and resulted in acceptable blastocyst development[3]. Administration of ionomycin (3mg/kg, i.p. injection, once daily) for 14 days improved hyperglycemia and insulin resistance, but reduced side effects compared with thiazolidinedione in a mouse model of diabetes. Ionomycin treatment also effectively inhibited cyclin-dependent kinase 5-mediated phosphorylation of PPARγ at Ser273 in vivo[4].
