KE00157 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The APOA1 ELISA kit is to be used to detect and quantify protein levels of endogenous APOA1. The assay recognizes human APOA1. An antibody specific for APOA1 has been pre-coated onto the microwells. The APOA1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody specific for APOA1 is added to detect the captured APOA1 protein. For signal development, Sterptravidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
APOA1 (apolipoprotein A1) is a plasma protein and a major protein component of high density lipoproteins (HDL) which is associated with reversed cholesterol transport, lipid/cholesterol binding, lecithin/cholesterol acyltransferase (LCAT) activation and specific receptors binding. It is synthesized in the liver and small intestine. Defects of APOA1 cause low HDL level and systemic non-neuropathic amyloidosis. Serum concentration of APOA1 is inversely related to the risk of developing atherosclerosis. Alterations of APOA1 level also correlate to cancer and Alzheimer's Disease, thus measurement of APOA1 can be used in diagnosis or prognosis of related diseases.
