KE00266 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The SFRP1 ELISA kit is to be used to detect and quantify protein levels of endogenous SFRP1. The assay recognizes human SFRP1. An antibody specific for human SFRP1 has been pre-coated onto the microwells. The human SFRP1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another horseradish peroxidase (HRP)-conjugated antibody specific for human SFRP1 is added to detect the captured human SFRP1 protein. For signal development, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Secreted Frizzled Related Protein 1 (SFRP1) belongs to the SFRP family that contains a cysteine-rich domain homologous to the putative Wnt-binding site of Frizzled proteins. SFRP1 has been classified as a tumor suppressor gene due to the loss of expression in various human cancers, which is mainly attributed by epigenetic inactivation via DNA methylation or transcriptional silencing by microRNAs. SFRP1 is a biomarker for aggressive subgroups of human gastric cancer and loss of SFRP1 expression is associated with poor prognosis in hepatocellular carcinoma. Patients with high SFRP1 expression in tumor tissue had a significantly better overall survival rate, and SFRP1 can be used as a prognostic biomarker for patients with poor survival.
