KE00198 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The TIMP4 ELISA kit is to be used to detect and quantify protein levels of endogenous TIMP4. The assay recognizes human TIMP4. An antibody specific for TIMP4 has been pre-coated onto the microwells. The TIMP4 protein in samples is captured by the coated antibody after incubation.Following extensive washing, another antibody of biotinylated specific for TIMP4 is added to detect the captured TIMP4 protein. For signal development, Sterptravidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Endogenous inhibitors of matrix metalloproteinases (MMPs) called tissue inhibitors of matrix metalloproteinases (TIMPs) form a family of four members,TIMP1-4. Although possessing many similarities, TIMPs show several structural and biochemical differences, and their expression patterns differ. TIMP4 is characterized by being primarily restricted to cardiovascular structures. Experimental studies have suggested a role for TIMP4 in cardiovascular disorders exhibiting an inflammatory component. Overexpression of TIMP4 has been observed within the arterial wall in rats after a balloon injury. TIMP4 has been shown to suppress vascular smooth muscle cell migration and induce cell apoptosis and to inhibit capillary endothelial cell migration.
