KE00249 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The Thrombospondin 1 ELISA kit is to be used to detect and quantify protein levels of endogenous Thrombospondin 1 . The assay recognizes human Thrombospondin 1 . An antibody specific for Thrombospondin 1 has been pre-coated onto the microwells. The Thrombospondin 1 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for human Thrombospondin 1 is added to detect the captured human Thrombospondin 1 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information
Thrombospondin 1(TSP-1), an extracellular matrix protein, is the first identified natural angiogenesis inhibitor. A variety of normal cells, including endothelial cells, fibroblasts, adipocytes, smooth muscle cells, monocytes, macrophages, and transformed cells such as malignant glioma cells, secrete Thrombospondin 1. It regulates cellular phenotype during tissue genesis and repair. It acts as a molecular facilitator by bringing together cytokines, growth factors, matrix components, membrane receptors and extracellular proteases. Thrombospondin 1 binds to a wide variety of integrin and non-integrin cell surface receptors. It is also a major activator of transforming growth factor (TGFβ1).
